Modern Mariana Islander DNA is difficult to work with because of their diverse sources of admixture, many cases in fairly large amounts. They all have European admixture, which is not too surprising since the islands have been ruled by Spain, Germany, or the United States for roughly the past four centuries. Perhaps more unexpected (but still easily explicable historically) is their Indigenous American admixture; a lot of the "Spanish" admixture in Pacific islands, including the Marianas and the Philippines, is actually from New Spain and therefore is accompanied by a great deal of Indigenous American DNA, as these "Spaniards" in fact have been mestizos like most present-day Mexicans.

A more difficult confound is the recent Asian admixture in Mariana Islanders. Many of them have recent ancestry from the Philippines (which may be the source of the prehistoric founders of the indigenous population of the Mariana Islands) and Japan (which has included Korea and Taiwan at the time of emigration of some of these individuals' ancestors).

I suppose that the most likely source for Native Oceanian admixture in present-day inhabitants of the Mariana Islands should be Carolinians, who are a group of Oceanic Austronesians who have migrated to the Marianas during the historical era from further south in Micronesia.

The movements of ancient humans can be difficult to ascertain from their current population genetic structure. Studying the peopling of the Micronesian islands, Liu et al. examined 164 ancient human remains from five different archaeological sites in remote Oceania from different prehistoric time frames, along with 112 present-day individuals from the same area. They combined these new data with the results of previous studies and also compared their results with linguistic studies. Their analysis revealed successive movements from island Southeast Asia that differ from those in the southwest Pacific.
